Breakage and condensation of a stretched lacO DNA filament
LacO between two centromeres in anaphase. The lac operators are evenly
spaced along the 10kb of the lac operator array. When the arrays are stretched
in vivo, alternating regions of bound (fluorescent) and unbound (not fluorescent)
chromatin can be visualized. As shown in the time-lapse, bound and unbound
domains can be delineated. These fluctuations reflect heterogeneity in
lac-repressor binding that become apparent in analysis of single molecules.
